The contribution of saponin hemolysis in observing membrane properties of erythrocytes

Volume changes in human erythrocytes by saponin were determined during hemolysis. The erythrocyte volume increased after a lag time, reached a maximal volume, and then the erythrocytes hemolyzed. There was a close correlation between the reciprocal of the time and the saponin concentration. Hemolysis was inhibited by aldonolactones. A strong correlation existed between the maximal volume in saponin hemolysis and the critical volume in hypotonic hemolysis. A strong correlation also was observed between the lag time and the cholesterol concentration. This approach to saponin hemolysis should be useful in the study of susceptibility of erythrocyte membrane to saponin.
