Effect of uptake blockers on 5-hydroxytryptamine & noradrenaline induced responses on rat anococcygeus muscle

5-Hydroxytryptamine (5-HT) and noradrenaline (NA) produced dose-dependent contractions of rat anococcygeus muscle. The responses to 5-HT were inhibited significantly by cocaine (4.3 .times. 10-6 M), imipramine (1.0 .times. 10-6 M) and dexamethasone (9.3 .times. 10-5 M). All these drugs potentiated the responses to NA. The graded doses of imipiramine produced dose dependent inhibition of the responses to 5-HT. The antagonism was noncompetitive. Cocaine in a higher dose produced a dose-dependent contraction of rat anococcygeus muscle. The per se effect of cocaine was blocked by phenotolamine. Indirect sympathomimetic effect of 5-HT and cocaine involving release of NA is suggested.
