Experiments in the freezing and storage of equine embryos

Embryos, recovered non-surgically 6 and 8 days after ovulation, were cooled to 0 deg , -10 deg , -15 deg or -20 deg C, or stored at 20 deg for 8 min to 9.2 h, -5 deg for 10 min, or -196 deg for 1-30 days. Dulbecco's phosphate-buffered saline containing antibiotics (PBS) and PBS plus 20% mare serum (PBS + S) were used for day-8 and day-6 embryos resp. None of the day-8 embryos that were stored at -196 deg in the presence of 1.5 M DMSO or 1.0 M glycerol developed in recipient mares. Half of the day-8 embryos stored at 20 deg for 8 min to 9.2 h and half of those cooled to 0 deg in DMSO-free PBS survived. None of the day-8 embryos and 1 day-6 embryo developed after storage with 1.5 M DMSO at 20 deg . One day-6 embryo survived cooling to at least -10 deg C in 1.5 M DMSO. Three of 11 day-6 embryos stored at -196 deg in 1.0 M glycerol developed, but day-6 embryos similarly stored in 1.5 M DMSO did not survive.
