NK-2.1: an NK-associated antigen detected with NZB anti-BALB/c serum

NZB/B1NJ mice immunized with BALB/c spleen cells targets. To determine if NZB anti-BALB/c antibodies recognized an allele of the previously described locus NK-1, 136 (BALB/c .times. NZB)F2 progeny were screened for sensitivity to NZB anti-BALB/c serum and anti-NK 1.1 (BALB/c .times. C3H F1 anti-CE serum) and C. Of the F2 progeny, .apprx. 1/4 reacted like the BALB/c parent and 1/4 like the NZB/B1NJ parent; the occurrence of F2 mice that were nonreactive with either antiserum or were strongly reactive with both suggested that the antigens detected were nonallelic. Segregation analysis of antisera sensitivity and coat color phenotype of 63 (BALB/c .times. NZB)F2 indicated that sensitivity to NZB anti-BALB/c serum was linked to the agouti (a) locus on chromosome 2. Linkage of NK-1 to chromosomes 2, 4 or 7 was excluded. On the basis of the antigen detected by NZB anti-BALB/c serum is designated as NK-2 and the activity of the serum as anti-NK-2.1. Strains (18) were tested for reactivity with anti-NK 2.1 and anti-NK-1.1. Nine were NK-1.1+, NK-2.1-; 6 were NK-1.1-, NK-2.1+. C57BL/6 and DBA/1 were NK-1.1+, NK-2.1+; 129 was NK-1.1-, NK-2.1-. Quantitative differences in sensitivity to C-dependent lysis with the 2 antisera were noted among the 18 strains as well as among F1 and F2 progeny between NK-1.1+ and NK-2.1+ parental strains. SC of C57BL/6 and DBA/1 that are low NK-2.1+ were labeled with anti-NK 2.1 serum and fluoresceinated goat anti-mouse Ig. NK-2.1+ cells, selected with the fluorescence-activated cell sorter, were enriched in NK activity compared to NK-2.1- SC.
