Ionic exchange column chromatography on SP-Sephadex C-50 followed by gel filtration on Sephadex G-75 revealed aging effects that occurred in the .gamma.-crystallins of the calf lens cortex and nucleus. The .gamma.-cryatallins of the calf lens cortex and nucleus showed different relative quantities of major peaks II, III and IV. These 3 major peaks of the calf lens nucleus showed similar relative quantities. The calf lens cortex .gamma.-crystallins had prominent major peak of fraction II followed by III and IV. The minor protein fractions of the calf lens cortex had more bound carbohydrate than the nucleus. Analyses of polyacrylamide gel electrophoresis with Comassie and PAS [periodic acid Schiff] staining, absorption spectra and relative quantity studies further characterized the minor .gamma.-crystallin fractions associated with the carbohydrate.
