Demonstration of gamma irradiation induced synthesis of mRNA in potato buds

After gamma irradiation, there was a 2- to 3-fold increase in RNA synthesis which was sensitive to actinomycin D in potato bud tissue. The poly(A) containing RNA was separated from the total RNA synthesized during this phase by affinity chromatography on poly(U) sepharose. About 1% of the total RNA synthesized was polyadenylated. Non-poly(A) RNA was separated into 11 bands on electrophoresis and a major portion of the radioactivity was found in 4 bands. Electrophoresis of poly(A) RNA indicated that its size was in the range 12S and 4S and the poly(A) fragment isolated from the RNA exhibited sedimentation behaviour between 5S and 4S. Cordycepin inhibited poly(A) RNA synthesis.
