Cytoplasmic estrogen receptor (ER) from immature rat uterus and lactating rat mammary gland binds to heparin Sepharose. Mammary tumor ER from various origins displays a low binding ability to the matrix (tumors: DMBA of the mouse; MCF-7 cell line and human breast cancers). This low binding ability may be due to the catalytic action of an enzyme, which rapidly alters the heparin binding site of the receptor. Attempts to stabilize the binding of mammary tumor ER to heparin-Sepharose were unsuccessful.
