Response of human leucocytes to yellow fever virus infection in vitro

The response of J-111 cells (derived from human leukemic leukocytes) to yellow fever virus (YFV) was investigated. Although the cytopathic effect (CPE) was not clear, the YFV-infected cells showed a significant increase of Feulgen-positive substances (FPS) in the nuclei. In the same cell cultures infected with dengue virus (DV, type 1), the FPS increased similarly but amounts were less than those seen in the YFV-infected cultures. The DNA content of FPS-containing cells was measured by a microspectrophotometer. While the DNA decreased during the course of incubation both in infected and non-infected cultures, YFV-infected cell DNA increased transitorily 4-5 days after virus inoculation. Apparently the YFV infection caused a shift of DNA in the human leukocytes and the response was more intense than that seen in the same cells infected with DV.
