Mutagenic activity of isoniazid & its metabolites in different test systems

Possible mutagenic effects of Isoniazid (INH) and its metabolites are evaluated using the Salmonella mammalian microsome assay and 2 other mammalian in vivo tests, namely dominant lethal test and mouse testicular DNA synthesis inhibition test. Isoniazid was mutagenic in 2 of the 4 strains tested by Ames method (TA 100 and TA 1535) in absence of the microsomal fraction (S9). Of the hydrazine metabolites, hydrazine sulfate (HS) and monoacetyl hydrazine (MAH) were mutagenic. Antitoxicants of INH did not show dose dependent inhibition of mutagenic activity of INH. The response of INH and hydrazine was negative in dominant lethal test, whereas DNA synthesis in mouse testis was inhibited by INH, HS, MAH and acetyl INH.
