Continuous in vitro cultivation of Plasmodium falciparum: replacement of human serum by calf serum and proteose peptone

A continuous in vitro culture experiment was performed to compare the effects of 10% human serum, 10% calf serum supplemented with 5% human red cell extract fraction I, and 10% calf serum supplemented with proteose peptone (0.25 ml of 15% stock solution) on the growth and multiplication of P. falciparum. In the 3 respective culture systems the average parasitaemia on day 0 was 1.99, 1.66 and 1.87% and on day 7 was 27.5, 24.9 and 22.8%. These results indicate that calf serum and proteose peptone (both commercially available and relatively inexpensive) provide a satisfactory substitute for human serum in P. falciparum cultures.
