Improvement of technical processes for isolating preparations of animal glomeruli of highly uniform size and free from an tubular admixture is discussed. A high degree of homogeneity is desired to show minute variations in diameter of flocculi after incubation with various mediators or pharmacologic reagents. Superficial and juxtamedullar glomeruli are of different size. Most of the experiments were done on rats, but some on pigs, which have very voluminous kidneys. Four different techniques of isolation were proposed: injection of colloidal Fe after which golomeruli, having absorbed Fe particles, can be attracted by a magnet, dissection of the nephron after enzymatic digestion, centrifugation in a Ficoll gradient. A sieving technique, previously used in glomeruli of rats and rabbits. Four combinations of 2 superimposed screens were compared.
