Covalently closed circular 3 .mu.m DNA is an extrachromosomal copy of S. cerevisiae ribosomal DNA. HindIII digestion of 3 .mu.m DNA produces 2 fragments of 6300 and 2600 base pairs. Both fragments were cloned in pBR322 cloning vehicle. Restriction analysis of recombinant plasmids was used to design a physical map of 3 .mu.m DNA.
