Ornithine decarboxylase (EC 4.1.1.17) of E. carotovora 438 is a very labile enzyme which is inactivated when cells are treated and decomposed by chemical, mechanical, physical and biological methods. Pyridoxal phosphate and dithiothreitol added to the homogenate did not restore ornithine decarboxylase activity. Cell decomposition in the presence of cofactor and dithiothreitol allowed isolation of a homogenate whose activity per protein unit was higher than that in intact cells. The ornithine decarboxylase activity is not connected with structural organelles of the cell and is incorporated into a soluble cell fraction.
