Effect of monocrotophos on the neurotransmitter system in the central nervous system

The effect of 1 dose of monocrotophos at 0.1, 0.05 and 0.025 LD50 was observed during 24 h. In another investigation 0.05 LD50 was given daily for 30 days to white rats intragastrically by gastric tube. The effects of monocrotophos on the metabolism of catecholamines and serotonin, glutamic acid decarboxylation and cholinesterase activity in rat brain were studied. Cholinesterase activity was reduced in whole blood and brain after 1 dose at 0.1 and 0.05 LD50 and after 30 days of poisoning with 0.05 LD50. In acute poisoning, the level of norepinephrine decreased. In long-term poisoning epinephrine level and MAO activity were reduced. No effect of monocrotophos was observed on glutamic acid decarboxylation and levels of serotonin and 5-hydroxyindolacetic acid. Levels of serotonin precursors, free Trp and 5-hydroxytryptophan, increased. The toxic action of monocrotophos depended on inhibition of cholinesterase activity with raised acetylcholine level and disturbances in the equilibrium of many neurotransmitters in the CNS. The adrenergic system was most sensitive to this toxic action.
