Male guineapigs weighing 190 to 250 g were given a diet free from vitamins A and C without or with retinyl palmitate 5000 IU/kg diet, ascorbic acid 2 mg/ml drinking water or both. After 16 days they were killed. The post-mitochondrial supernatant fraction from the liver was incubated with aflatoxin B1. Preparations from guineapigs deprived of vitamins A and C metabolized significantly less aflatoxin B1 and produced less of the metabolite AFII than those of controls. Production of AFI was increased by deficiency of vitamins A and C but decreased by deficiency of vitamin C alone. Production of aflatoxin M1 was not changed by any diet.
