Effects of reduced glutathione in endotoxin shock--change of blood chemical mediators

The anti-toxic effect of reduced glutathione (GSH) in endotoxin shock was studied. Rabbits were anesthesized with i.v. pentobarbital sodium. The femoral artery was cannulated for recording blood pressure and collecting blood. GSH from 100-1000 mg/kg was administered in single i.v. injections 15 min prior to endotoxin infusion. Blood samples for histamine and serotonin determination were obtained 1, 3, 5, 15, 60 and 120 min after endotoxin injection and the results were read on a fluorometer using the method of Shore and Udenfriend, respectively. The blood concentration of chemical mediators after endotoxin administration was elevated. The elevation of plasma histamine concentration was blocked by the administration of GSH 500 mg/kg. The elevation of plasma serotonin concentration was blocked slightly by the administration of GSH 500 mg/kg. The reduced glutathione inhibited the release of chemical mediators, preventing endotoxin shock.
