Several bisamidines of 2,6-diaminoanthraquinone were about as potent as metronidazole against experimental caecal amoebiasis in rats. The safety margins were somewhat lower, but since the drugs did not contain a nitro-moiety, they were sufficiently interesting to warrant further consideration. Against experimental hepatic amoebiasis in hamsters these drugs were, in general, more potent and had a greater safety margin than metronidazole. (2,6-anthraquinonylene)bis[N',N"'-diethylacetamidine], selected for additional studies, was highly effective for the treatment of experimental amoebiasis in rats and hamsters; the safety margins compared favourably with those of metronidazole. The absence of a nitro group in the molecule, the lack of mutagenicity in the tests, and the high degree of activity make this series of compounds interesting as anti-amoebic agents and further evaluation is in progress.
