Lipogenetic effects of ethanol and ethanol metabolism in the rat pancreas in vitro

Pancreas from rats was incubated in a medium with 10 mM [U-14C]glucose, 0.5 mM [1-14C]acetate, 0.5 mM [1-14C]palmitate, [3H]water and [1-14C]ethanol. With 10 mM ethanol, oxidation of [1-14C]acetate and [1-14C]palmitate and uptake of [U-14C]glucose were decreased by 15, 20 and 20%, respectively. The proportion of 14C from [1-14C]acetate recovered in glyceride palmitic acid increased at the expense of longer and unsaturated glyceride fatty acids, suggesting specific stimulation of the malonic pathway. With ethanol 100 to 200 mM, oxidation of [1-14C]acetate and [1-14C]palmitate decreased by 25 to 45%. Lipogenesis from [3H]water and [1-14C]acetate was increased 2.5- and 2.0-fold and proportion of 14C in glyceride palmitic acid increased. The esterification of [1-14C]palmitate into triglycerides and phosphatidic acids increased by 45 and 90%. The metabolism of 0.5 mM [1-14C]ethanol was compared with that of an equimolar concentration of [1-14C]acetate. The oxidation of [1-14C]ethanol and [1-14C]acetate to 14CO2 was 8 and 273 nmol/mg wet weight h, respectively; the incorporation into glyceride fatty acids was 0.4 and 8.1 nmol/100 mg. The rate of oxidation and of lipogenesis from [1-14C]ethanol increased with increasing concentration from 0.5 to 100 mM. At 10 mM, [1-14C]ethanol provided 26% of the carbon used in lipogenesis.
