Gel filtration and ion-exchange chromatography techniques for purification of crude Theileria antigen obtained from infected calf erythrocytes are outlined. The suitability of resulting antigen fractions for erythrocyte sensitization was compared, employing Ouchterlony precipitation, passive haemagglutination, quantitative protein and carbohydrate determination and immunofluorescence techniques. It is proposed to fix fraction 2 (of fraction P-1) on bentonite or latex particles for use in the agglutination test, instead of the passive haemagglutination test.
