Dose and time relationships in manganese-bilirubin cholestasis

Combination of noncholestatic doses of Mn and bilirubin produced a dramatic fall in bile flow in rats that was dose dependent for bilirubin but not for Mn. The dose of Mn linearly influenced the period of time during which Mn bilirubin (Mn-BR) cholestasis could be elicited. The protection afforded by sulfobromophthalein (BSP) was also studied. The magnitude of the protection seemed related to the amount of BSP remaining unexcreted at the time the bilirubin was injected. The relationship between the minimal protective dose of BSP and the dose of bilirubin was linear and independent of Mn load. BSP appears to act on the bilirubin component of Mn-BR cholestasis, rather than on Mn.
