The effect of the pituitary factor (PF) isolated from sheep pituitary glands on the cell size distribution pattern of thymocytes was studied using a Coulter Counter. It induces an enlargement of cell volume in isolated thymocytes . A procedure for the quantification of this effect of PF is described. The effect of PF is dose-dependent and is observed within half an hour of incubation. Maximum effect was obtained on thymocytes though a significant effect was also observed on cell derived from spleen and bone marrow. PF did not induce immunocompetence in thymocytes as measured by their graft vs. host reactivity in chick embryo system. The effect of PF on cell size could not be due to maturation of these cells. The swelling of cells may be related to the triggering of these cells to divide by PF.
