The biuret, ultraviolet (NAR 27, 1387) and Lowry (NAR 21, 4458) methods for the estimation of total protein content were compared in 31 samples of serum and in 30 of rat heart homogenates. There were significant differences in values. Values were lowest with the Lowry method and, for serum and homogenate, were highest with the ultraviolet and biuret method, respectively. Possible sources of these discrepancies are discussed. Although the biuret method was least sensitive it was the most reproducible.
