The effect of bovine pancreatic ribonuclease (A RNase) and bull seminal ribonuclease (AS RNase) on human lymphocytes undergoing blastic transformation

A RNase and AS RNase had stimulatory effects on PHA incorporation. In human lymphocyte cultures, during PHA-induced blastic transformation, the dose of 100 .mu.g of A RNase/ml medium had a slight inhibitory effect; the same dose of AS RNase had a marked inhibitory effect. Lymphocytes from different individuals exhibited individual reactivity. The kinetics of the effect of both ribonucleases on human lymphocyte cultures was studied from 48-192 h with a dose of 100 .mu.g/ml medium. During this time the incorporation of 3H-TdR was significantly reduced in the cultures treated with AS RNase, whereas it was increased in the culture treated with A RNase. Thus, AS RNase had a marked inhibitory effect on human lymphocyte cultures stimulated by PHA for blastic transformation, and A RNase showed a stimulatory effect. Indirect immunofluorescence demonstrated that none of the RNase was bound to the lymphocyte membranes in normal PHA-unstimulated human peripheral blood lymphocytes. However, lymphocytes undergoing PHA-induced blastic transformation exhibited strong fluorescence when incubated with AS RNase, and very poor fluorescence when incubated with A RNase.
