Repercussions on hepatic lipogenesis in the rat during realimentation with diets of different protein content

To study the relationship between protein diets and liver lipogenesis, male Wistar rats were 1st put on a normal protein diet, then on diets with varying amounts of protein. The animals were sacrificed and spectrophotometric assays of various enzymes in liver samples were done. The changes in 6 enzyme parameters in rat liver determined during the fasting period before the change in diet, and during subsequent normoprotein, 0 protein and hyperprotein diets are listed. A normal protein diet increased all liver enzymes tested, while the preliminary fasting period depressed them, as did a non-protein diet. A non-protein or a hyperprotein diet inhibited lipogenesis and lipogenic enzymes, with the exception of acetyl-CoA carboxylase. This enzyme and fatty acid synthetase are the limiting enzymes of lipogenesis. The concentration of of acyl-CoA, noted inhibitor of lipogenesis, diminished as the protein supply was reduced.
