Sialyl oligosaccharides from milk

Sialyl oligosaccharides were prepared from human milk by Sephadex G-25 gel filtration, deproteinization with ZnSO4 neutralization with Ba(OH)2, centrifugation of the chilled suspension, and lyophilization of the supernanant to give a yield of 1.2 g mixed sialyl oligosaccharides/l milk. The oligosaccharides were separated by dissolving the fraction in 2 mM acetic acid/pyridine buffer (pH 5.4), applying it to a DEAE cellulose column, and eluting with 12 mM and 60 mM acetic acid/pyridine buffer (pH 5.4). The sialyl oligosaccharides were identified by comparing their electrophoretic and chromatographic mobilities with those of standard oligosaccharides.
