Brain tissue sections, homogenates and mitochondria were prepared and incubated in different media. In Krebs-Ringer phosphate medium with 5 mM (normal) K, 10-4 and 10-5 M of amphetamine caused an increase in O2 consumption and no difference in glucose. This trend continued at lower amphetamine concentrations if Ca was removed from the medium. If K is increased and the amphetamine level raised to 10-3 M, O2 and glucose consumption dropped. In tissue homogenate O2 consumption increases at 10-4 and 10-5 M of amphetamine in the presence of substrates and cofactors. Mitochondrial oxidative phosphorylation was not significantly modified by comparison with controls.
