A series of compounds related to ethyl 2-(4-dibenzofuranyloxy)-2-methylpropionate (1) and clofibrate with variations in the link between the aryl and the isobutyric acid parts were investigated for lipid-lowering effect in spontaneously hyperlipidemic rats. Compounds in the 2 series, in which the group connecting the aryl moiety to the isobutyrate part was able influence of an .alpha.-oxygen in the short-chain compounds was shown, whereas the chain length was less critical for activity. Compound 1 is less sensitive than clofibrate to alterations in the side chain. Three compounds with 2 or 3 C units inserted in the side chain of clofibrate were inactive. Three compounds related to ethyl 4-chlorophenylacetate (12) with a hydroxyl or a methoxyl group in the .alpha.-position were found to possess triglyceride-lowering properties in contrast to the inactive compound 12. The positive influence of an .alpha.-oxygen was thus confirmed also in this series of compounds.
