Potential plasma lipid-lowering (12) compounds belonging to the 2-(dibenzothiophenoxy)-2-methylpropionate series were studied. They were all prepared through the reaction of the appropriate dibenzothiophene-ol with 1,1,1-trichloro-2-methyl-2-propanol under acidic conditions, followed by esterification. In connection with the synthesis of various dibenzothiophene-ols, metalation and halogen-metal interconversion of various dibenzothiophene-ols was extensively utilized. The 2-(dibenzothiophenoxy)-2-methylpropionates constitute a new class of hypolipidemic agents with a structure-activity relationship resembling that of the 2-(dibenzofuranyloxy)-2-methylpropionates. Optimum activity is obtained when the side-chain is situated in the 4-position. Studies were done in spontaneously hyperlipidemic rats.
