Simplified thin-layer chromatographic method for the stimultaneous determination of clonazepam, diazepam and their metabolites in serum

When epileptic patients are admitted as emergency cases, they are usually given diazepam, and if the desired response is not observed, then clonazepam is often administered. This clinical procedure necessitates a method for simultaneous and quantitative measurement of the 2 drugs and their metabolites. A method in which the diffuse light reflectance of both substances and their metabolites, which have been simultaneously extracted and separated, are measured directly on an unstained TLC plate is presented. This quantitative method is rapid and precise. Results are well within accepted limits of deviation. Five metabolites (N-desmethyldiazepam, 3-hydroxydiazepam, oxazepam, 7-aminoclonazepam and 7-acetamidoclonazepam) were also recovered from serum by this method.
