Included in this review are sections on cellular and tissue specificity of oxytocin binding sites, binding affinity and ligand specificity, oxytocin binding by isolated mammary cells, and metal ion requirements for oxytocin activity and binding activity. The binding activity has been localized in enriched plasma membrane fractions from the rat mammary gland. Apparent dissociation constant for interaction of oxytocin with its receptors in various tissue preparations is in the nanomolar range. The affinity of binding sites for oxytocin analogues corresponds generally to the potencies of the analogues as agonists or antagonists.
