RNA concentration in DNA preparations from animal tissues (quantitative analysis of an alkaline hydrolysate)

Two methods are compared for estimating the RNA admixture in preparations of DNA isolated from mammalian tissues. The RNA content in alkaline hydrolyzates of DNA as determined by the spectrophotometric method is found to be from 2 to 5 times higher than that estimated by means of the colour orcinol reaction. Possible reasons of such a difference are discussed. Attempts are made to find out the optimal conditions for isolating RNA from DNA preparations.
