Mechanism of action of antitumor antibiotic mithramycin

The effect of mithramycin, an antitumor antibiotic, was studied at in vivo, cellular and nuclear levels. The antibiotic inhibits RNA synthesis in vivo in YAS [Yoshida ascites sarcoma] and to a lesser extent in rat liver. The concentration required for bringing 50% transcription inhibition in isolated nuclei (10-4 M) from YAS cells and rat liver was higher than that required for isolated cells (10-6 M) from YAS cells and rat liver. The significance of these results was discussed in the light of the mechanism of action of this antibiotic.
