Yersinia enterocolitica. I. Isolation of antigenic fractions from Y. enterocolitica

To clarify the cellular components of Y. enterocolitica related to toxicity and antigenicity, bacterial cell saline (Fr. I), hot phenol-water extracts (Fr. II) and sugar components (Fr. III) from hydrolysis of Fr. II with 0.1 N acetic acid were studied. TLC and liquid chromatography showed the sugar composition to be essentially the same among Fr. I, II, and III: galactose, glucose, xylose, and an unidentified sugar, possibly 6-deoxy-L-altrose. In the double gel-diffusion precipitation test against anti-Y. enterocolitica whole cell antiserum, Fr. I, II, and III formed 1 fused precipitation line, and Fr. I and II an additional precipitation line. Even heating at 100.degree. C for 1 h or treatment with chloroform did not alter the precipitation pattern of Fr. I. When 250 .mu.g (as sugars) of Fr. II was injected i.v. into 5 mice, 2 mice died within 72 h; all 5 mice injected with 1500 .mu.g of Fr. II died. No mice died when injected i.v. with 2000 .mu.g of Fr. III.
