Six different polyvinyl alcohol polymers ranging in molecular weight from 35,000 to 240,000 were compared for their pathologic properties in rats. They all caused anemia and were found to infiltrate various organs and tissues. Four caused hepalosplenomegaly. The two that failed to enlarge the spleen nonetheless caused striking histologic changes in these organs. All of them caused thymus involution, but none induced marked adrenal hypertrophy. Osly two polymers proved to cause hypertension and widespread vascular lesions, hypertrophy of the heart and kidneys, extensive renal pathology, increased thyroid weight, enhancement of hepatosplenomegaly, anemia, and thymic involution. One of the two caused edema, ascites, a high mortality rate, and changes reminiscent of eclamptogenic toxemia. Insofar as hypertension and the associated changes are concerned, it is suggested that a relative cold water insolubility is the one physicochemical characteristic that appears to be associated with a high degree of pathogenicity. There is no relationship between any detectable effect and the molecular weight.
