The influence of folic acid on the anticonvulsive action of phenobarbital and diphenylhydantoin

In mice of Porton strain phenobarbital 25 mg/kg and diphenylhydantoin 20 mg/kg alone or with folic acid 100 mu g/kg in 1% methylcellulose solution were introduced into the stomach and, 30 min later, convulsions were induced by subcutaneous injection of strychnine nitrate solution 1 mg/kg or pentamethyltetrazol 85 mg/kg. Folic acid did not affect the anticonvulsant activity of phenobarbital, but that of diphenylhydantoin on the induction of pentamethyltetrazol fits was reduced when the compounds were given together every second day for 3 weeks.Wistar rats were given by mouth 1.5 mu g folic acid/g alone or with phenobarbital 25 mg/kg or diphenylhydantoin 200 mg/kg, and folic acid activity was estimated with Lactobacillus casei in blood samples taken 30, 45 and 60 min later. Diphenylhydantoin reduced folic acid activity in serum, but the phenobarbital and folic acid there was only a slight decrease at 60 min.
