The optimum methods of creating and treating a depot of radioactivity that would make a dose of injected 241Am available to the circulation at a relatively slow rate was investigated. Total and partial body counting determined the distribution of 241Am within the body of beagles injected with 241Am, and dogs injected with 241Am and Zn-DTPA. Untreated dogs showed retention similar to a phantom, while those treated with Zn-DTPA retained significantly less. Daily injections of Zn-DTPA accelerated 241Am disappearance from a simulated wound site, prevented its translocation from the depot to the liver and skeleton and reduced total body 241Am content through increased secretion. Administration of Zn-DTPA over a 5 period interval/day proved more effective than administration of the same dosage given in one daily injection.
