Brain .alpha.-aminoadipic acid .delta.-semialdehyde was analyzed by 2 methods. The 1st method consisted of performic acid oxidation of myelin protein to convert the aldehyde to .alpha.-aminoadipic acid residue and hydrolysis of the oxidized sample followed by densitometric assay of an electrophoretogram. The other method was the detection of .epsilon.-hydroxynorleucine in the alkaline hydrolysate of 3H-NaBH4 reduced preparation. .alpha.-Aminoadipic acid .delta.-semialdehyde was present in myelin proteins in addition to the previously demonstrated sources: collagen, elastin, histone and erythrocyte membrane.
