There is now considerable interest in the construction of plasmids containing Dna fragments from a variety of bacterial genera, for purposes of studying the expression of the genetic potentialities of the incorporated bacterial Dna in different bacterial hosts. We report that the self-transmissible plasmid RP4 has only one site susceptible to cutting by the enzyme Eco RI, and should therefore be a very suitable bacterial Dna vector in such experiments.
