Bovine semen was centrifuged at 7000 g and 0 deg -4 deg C for 10 min, the sperm pellet was resuspended in chilled water and recentrifuged, and the packed sperm mass was homogenised by resuspension and centrifuging at 10 000 revolutions/min for 20 min. The homogenised material (fraction A) was layered in 0.75 M sucrose and centrifuged; the supernatant (fraction B) was decanted and the sediment layered in 1.5 M sucrose and centrifuged (fraction C, supernatant; fraction D, sediment). Fraction A contained separated heads, midpieces and tails; fractions B, C and D were predominantly (90%) tails, midpieces and heads resp.
