5-Methylcytosine localised in mammalian constitutive heterochromatin

Use of an immunofluorescence technique showed that anti-deoxy-S-methylcytidilic acid (anti-5-MeC) binds preferentially to the C-band regions of centromeric heterochromatin in the mouse. The technique described can be used to locate classes of highly repetitive DNA which contain 5-MeC; these classes show remarkable quantitative variation between homologues and among individuals. Differences of this kind provide chromosome markers that can be used in genetic linkage studies or in the analysis of structurally altered chromosomes.
