Implant chemotherapy of experimental filariasis

The in vitro rate of diffusion per 24 hours of famphur (O,O-dimethyl O-p- phenylphosphorothioate) from polydimethyl-siloxane (PDS) capsules into human plasma was constant and was directly proportional to the capsule lumen surface area and inversely proportional to the capsule wall thickness. One capsule implanted subcutaneously into an experimentally infected jird almost completely eliminated microfilaraemia of Litomosoides carinii while in situ (5 weeks) but did not affect Dipetalonema wite microfilaraemia or adult worms of both species. The capsules were well tolerated by the jirds and little tissue response to them was observed.
