Colorimetric estimation of pantothenates in formulations

The method of Wollish and Schmall (Analytical Chemistry (1952), 29, 1509) was modified. The sample is heated with HCl and Zn at 90 deg C for about 5 min and made just alkaline with 50% NaOH. The filtrate and washings are made slighly acid with 0.1 N HCl and pyridine-cyanide reagent and ninhydrin solution are added. After heating at 85 deg to 90 deg for 10 min the solution is diluted and absorbance at 570 nm is measured. Pantothenate 2 to 8 mu g/ml can be estimated without interference from other vitamins, amino acids and tetracyclines.
