An automated Rose Bengal agglutination test using the ADAM system

An automated Rose Bengal agglutination test for brucellosis is described. Antigen is deposited in a discrete profile on to a moving transparent plastic tape and serum is added direct from blood-sampling tubes using a plastic reservoir device to maintain a constant serum level within the tubes. The serum/antigen mixture is stirred by air jets in an incubation chamber and the degree of agglutination read by a fixed optical scanning system. The results of 16836 tests carried out on the machine are presented. The sensitivity of the test, in relation to the serum agglutination test and complement fixation test, is about 92% and is better than the equivalent manual test. The specificity of the test is 89%, rather less than that of the manual test. The machine is capable of handling up to 1200 blood samples per hour and has a potential for use in other agglutination tests.
