Lipogenesis by in vitro liver and adipose tissue preparations from neonatal swine

The synthesis in vitro of lipid was examined in adipose tissue and liver preparations from pigs 2 to 60 days of age. The lipid content of adipose tissue and liver at various ages was estimated. In adipose tissue, the incorporation of glucose or acetate into lipid was low until weaning and thereafter significantly increased. Acetate was primarily incorporated into fatty acids at all ages, whereas glucose was equally incorporated into glyceride-glycerol and fatty acids before weaning, with a shift toward fatty acid as the primary product after weaning. The major class of lipids synthesized at all ages was triglyceride with either precursor. A number of enzymes associated with lipogenesis were examined and all activities increased considerably after weaning. Malic enzyme and citrate cleavage enzyme activities were low at birth and increased strikingly. The rate of synthesis of lipid by liver preparations with either glucose or acetate as precursor was limited compared with that of adipose tissue.
