Collectively, the in vivo data show that PGEs can promote osteogenesis in a variety of circumstances. The osteogenic response can include a stimulation of bone modeling and bone remodeling, depending on the bone envelope involved. Given systemically, PGEs promote osteogenesis and bone reconstitution in several animal models. Given locally, we have demonstrated that PGE, can promote site-directed osteogenesis. The ability to form new cementum, alveolar bone, periodontal ligament, and Sharpey's fibers insertions into the new bone in dog teeth near sites of local release of PGE,32 offers a novel and potentially very practical method for periodontal regeneration in conditions such as periodontal disease. The ability of locally applied PGE, to promote periodontal regeneration suggests that the same strategy might be useful in other clinical situations. PGs may enhance the ingrowth of bone and repair of soft tissues around dental and orthopedic implants. Because of its ability to promote connective tissue repair, perhaps by its ability to promote vascularization, it might also be used to promote the repair of ligaments and their reattachment to bone.
