Isolation and host range studies of Staphylococcus epidermidis and Micrococcus ssp. bacteriophage

One hundred four phage isolates were obtained from cultures of Staphylococcus epidermidis and Micrococcus spp. by 3 methods: Fisk cross culture technique, multiple-cross broth culture technique, and spontaneous lysis. The multiple-cross broth culture technique detected 31 phages that could not be isolated by the Fisk technique. The phages had 16 host ranges and lysed 75% of the S. epidermidis cultures but only 7% of the Micrococcus cultures. Phages isolated from 7 cultures of Micrococcus lysed only S. epidermidis cultures. The only phage isolated on a Micrococcus-propagating strain was from a lysogenic S. epidermidis culture and did not lyse any other Micrococcus. Six lysogenic S. epidermidis cultures were lysed by phages which had been isolated from them on other propagating strains.
