Simultaneous determination of three kinds of components in sanhuang tablets by high-performance liquid chromatography

An assay method for the simultaneous determination of 3 kinds of biologically active components and 5 compounds (emodin, chrysophanol, baicalin, wogonin and berberine hydrochloride) in Sanhuang tablets was developed using HPLC with a C18 column (150 mm x 4.6 mm ID, 5 micro m) set at 30 degrees C and acetonitrile as mobile phase (using gradient) with a flowing rate 1.00 mL/minute. The calibration curve of emodin was linear from 0.0207 to 0.207 micro g with r=0.999 9. The average recovery was 99.65% with 1.25% RSD. The calibration curve of chrysophanol was linear from 0.052 to 0.52 micro g with r=0.999 9 and an average recovery of 100.36% and RSD of 0.96%. The calibration curve of baicalin was linear from 0.2505 to 2.505 micro g with r=0.999 8 and average recovery of 100.22% and RSD of 1.29%. The calibration curve of wogonin was linear from 0.047 6 to 0.476 micro g with r=0.999 9, and an average recovery of 98.97% and RSD of 1.20%. The calibration curve of berberine hydrochloride was linear from 0.053 12 to 0.531 2 micro g with r=0.999 5, and an average recovery of 96.02% and RSD of 2.02%. The established method had also been used in the determination of the 5 compounds in 10 different batches of Sanhuang tablets. This method was proven to be accurate and quick, and can be used for the quality control of the herbal preparation.
