Fifteen patients with advanced malignant melanoma were treated with camptothecin at doses of 90-360 mg/m2 repeated every 2 weeks. None of the patients had a 50% or greater decrease in tumor diameter. Less pronounced transient tumor regression was noted in three patients, but no clinical benefit was associated with these responses. The remaining patients had no change or progression in their disease. Toxic effects included myelosuppression (11 patients), nausea and vomiting (9), alopecia (8), diarrhea (3), and hemorrhagic cystitis (1). We conclude that camptothecin, at least as administered in this study, has little to offer the patient with advanced disseminated melanoma.
