Pharmacological studies of a new anti-inflammatory drug, 1-phenylsulfonyl-5,5-diphenylhydantoin (PC-796) in experimental animals. I. Anti-inflammatory, analgesic and anti-pyretic activity

The anti-inflammatory, analgesic and anti-pyretic action of 1-phenylsulfony1-5,5-diphenylhydantoin (PC-796) were investigated using experimental animals. The following results were obtained. 1. PC-796 showed more potent inhibitory effects on the carrageenin, yeast and scald oedema in mice than did phenylbutazone and other non-steroidal agents. 2. PC-796 was found to possess inhibitory activity on the exudative reaction induced with croton oil in the granuloma pouch in rats. PC-796 showed also anti-exudative activity on the experimental peritonitis and pleuritis induced by carrageenin in rats. 3. PC-796 was found to be effective on the primary inflammation of adjuvant arthritis, more potent than acetylsalicylic acid and aminophenazone but slightly weaker than phenylbutazone. 1045 4. PC-796 showed the most potent inhibitory activity on the increased vascular permeability induced with bradykinin, serotonin, histamine, and hyaluronidase among the drugs tested. On further bradykinin actions, using the isolated ileum preparation in vitro, PC-796 was found to he effective at a concentration at which it did not show anti-serotonin and histamine activity, this activity of PC-796 was more potent than those of phenylbutazone and Aspirin. PC-796 also possessed inhibitory effect on the bradykinin-induced bronchoconstriction in guineapigs. 5. PC-796 was less active on the UV-erythema and had no effect on granuloma formation. 6. The inhibitory activity of PC-796 on the heat-denaturation of bovine serum albumin in vitro was found to be more potent than those of other drugs except for ibufenac. 7. The inhibitory effect of PC-796 on passive cutaneous anaphylaxis was more marked than that of acetylsalicylic acid, but less than that of phenylbutazone in rabbits. PC-796 showed more potent inhibitory effect on anaphylactic shock than did acetylsalicylic acid in mice. 8. Analgesic activity of PC-796 was mild, but the longest-lasting among non-steroidal anti-inflammatory agents tested in mice and rats. 9. PC-796 had more potent anti-pyretic activity than had phenylbutazone and acetylsalicylic acid in yeastfevered rats.
