Detection of human cytomegalovirus infection by FQ-PCR technique and its application in the diagnosis and treatment of HCMV infected children

Samples from 45 children suspected of having human cytomegalovirus (HCV) infection were examined using polymerase chain reaction (PCR), ELISA and fluorescent quantitative (FQ)-PCR. 25 HCV hepatitis cases were randomly assigned to receive ganciclovir combined with intravenous immunoglobulin (GCV + IVIG) or a control group. Patients from both groups were administered prednisone, glucurone, luminal and Xiaoyanlidanpian. The positive rates for PCR, ELISA and FQ-PCR were 60.0%, 33.33% and 66.67%, respectively, and their sensitivities were 84.38%, 46.88% and 93.75%, respectively. At baseline, there was no significant difference in viral DNA levels between the 2 treatment groups. Although viral load in both treatment groups decreased, the viral load in the group that received GCV + IVIG had more significant decreases (P<0.001). Results indicate that a viral level of 103 copies/ml detected using PQ-PCR can be taken as critical value for prediction of active HCV infection.
